摘要

对传统热水浸提取和超声辅助提取得到的鸡腿菇多糖AP1和CAP1的结构进行分析比较,研究了超声辅助提取对鸡腿菇多糖AP1结构的影响.结果表明:AP1由甘露糖、葡萄糖和半乳糖组成,CAP1由木糖、甘露糖、葡萄糖和半乳糖组成.红外光谱表明多糖AP1和CAP1在4 000~650 cm-1具有多糖类物质的特征吸收峰;不同的是,AP1的红外光谱中在865、764 cm-1处有吸收峰,而CAP1在903、826、756 cm-1处有吸收峰.原子力显微图中均显示多糖线形并具有分支结构.超声对其影响可能是引起了单糖之间连接的方式变化,进而更易散开形成较均匀的网状.

Abstract

The analyse of polysaccharides AP1 and CAP1 extracted from Shaggy Mane by boiling water and ultrasonic process is carried out.The affect of ultrasonic-assisted extraction on the structure of Shaggy Mane polysaccharide AP1 is investigated. The results showed that the components of Polysaccharides AP1 is identified by gas chromatography (GC) as: mannose,glucose,galactose;and CAP1 is xylose,mannose,glucose,galactose;the infrared spectra showed that polysaccharides AP1 and CAP1 have saccharide characteristic absorption peaks in the range from 4 000 to 650 cm-1.The infrared spectrum of AP1 has absorption peaks at 865 cm-1and 764 cm-1,but CAP1 has the peaks at 903 cm-1、 826 cm-1 and 756 cm-1.Atomic force microscopy (AFM) diagram showed the polysaccharides has branch structure.The ultrasound was likely to cause the connection pattern change of monosaccharides,and easily resulting in the formation of relatively uniform mesh structure.
